You probably don't have to drill holes through the PVC pipe but I did in my 135 mixed reef. The reason I chose to do so is the purpose of the PVC is to raise the LR off the DSB to prevent or delay dead areas under the LR. I thought, if you don't cut holes in the PVC there could be dead zones within the PVC pipe. So to allow flow through I drilled holes.
